HILIC

Development of separation analysis method using a column with complementary properties to ODS column
(HILIC, SFC, normal-phase mode)

ODS column in reverse-phase HPLC mode is generally used in purification and purity evaluation of Peptides, but a separation method is needed that has high orthogonality to ODS column since there is a possibility of impurity overlapping with the main peak.
To resolve this issue, PeptiStar has jointly developed a separation method that can be used for Peptide API manufacturing. It is now possible to separate impurities that were difficult to separate with conventional methods by using analysis column (DCpak P series) from Daicel Corporation in HILIC or SFC mode that enables separation that has high complementary properties to ODS column. PeptiStar utilizes either ODS column or DCPak P series in order to enable more accurate purity evaluation.

Advantages of HILIC Mode

  • Purification is possible in HILIC mode for compounds that are difficult to purify in reverse-phase system
  • Detailed analysis is possible when used in conjunction with reverse-phase system
  • High purity purification is possible by using fractioning purification technology that uses a combination of reverse-phase system and HILIC mode
